Abstract
Primers were designed based on the conserved regions at both ends of the alpha-gliadin gene coding region. PCR amplification was performed on the total DNA of the wheat variety “Liangmai No. 2” to obtain a DNA fragment of approximately 900 bp. The DNA fragment was cloned and sequenced to obtain three gene sequences. The genbank registration numbers are: dq417343, dq417344 and dq417345. Among them, dq417343 and dq417345 are 942bp and 921bp respectively, and can encode 313 and 306 amino acid residues respectively; while the length of the coding region of dq417344 is 852bp. Due to the presence of two premature stop codons, it cannot encode a functional mature protein and is a pseudogene. . Sequence alignment showed that these three genes were highly consistent with known ¦Á-gliadin genes, but there were some differences in the N-terminal repeat region and polyglutamine region.
