Cell Freeze-Thawing Assisted Hot Water Extraction Of Spirulina Polysaccharide And Optimization Of Deproteinization Process

Abstract
Using spirulina powder as raw material, the cell freeze-thawing technology and hot water extraction technology are combined to extract algal polysaccharide. Through single factor and orthogonal experiments, the extraction of remaining polysaccharides from broken cell residues after freeze-thaw and protein removal by Sevag method were studied, the process conditions were optimized, and compared with the traditional hot water extraction process. The results show that hot water extraction of broken cell residues has the best effect under the conditions of material-to-liquid ratio 1:16, extraction temperature 80¡ãC, and extraction time 1.5 hours. The total polysaccharide extraction rate after combining with cell supernatant Reaching 6.64%, which is nearly 14% higher than the traditional hot water extraction method; the optimal process conditions for protein removal using the Sevag method are an alkanol ratio of 5:1, a material-to-liquid reagent ratio of 3:1, and three treatments. Under these conditions, the protein removal rate and polysaccharide loss rate were 81.4% and 17.3% respectively. Further analysis of the crude polysaccharide quality showed that the total sugar content was 85.3%, the polysaccharide molecular mass was 84700u, and the polydispersity was 1.306; while the polysaccharide obtained by the traditional hot water extraction process had a molecular mass of 54800u and a polydispersity of 2.007, and the crude polysaccharide product quality was lower than Product quality obtained by cell freeze-thaw assisted hot water extraction process
