Abstract

In order to understand the complete sequence of the wheat PMH+ATPase gene and its related bioinformatics characteristics, the promoter of the wheat PMH+ATPase gene was cloned for the first time using the FullTail PCR method using the genomic DNA of Yumai 18, and cloned using the segmental cloning method. Its full-length sequence (accession number: AY829002). DNA sequence analysis results showed that the gene has a full length of 5770 bp and contains 15 exons and 14 introns. The first intron fragment is larger, 1432 bp long. Base component analysis results showed that the full length of the promoter and 5¡ä UTR of the gene were 161 and 201 bp respectively, and the G+C content was as high as 72.1% and 72.6% respectively. Analysis of its bioinformatics characteristics showed that the protein is composed of 951 amino acids, with a molecular weight of 104.6kD, 44 functional sites, 10 transmembrane regions, 1 CationATPase domain, 1 E1E2ATPase domain and 1 Hydrolase domain. Sequence comparison analysis with other plants shows that PMH+ATPase from different plant sources is highly conserved. The complexity of the structure of this gene and its putative product illustrates the precision of this gene in the regulation of life activities.
