Abstract
Abstract In order to construct a eukaryotic expression plasmid for herpes simplex virus type 2 (HSV-2)-infected cell polypeptide 27 (ICP27), PCR technology was used to amplify the ICP27 gene from the genome of HSV-2 333 strain and connect it to the eukaryotic expression vector pEGFPC2 , After performing colony PCR, enzyme digestion and sequencing identification on the positive clones, the recombinant plasmid pEGFPC2-ICP27 was successfully constructed. The recombinant plasmid pEGFPC2-ICP27 was transfected into Vero cells using Xfect transfection kit, and its expression was detected by RT-PCR and Western blotting. The results showed that the ICP27 gene was correctly expressed in Vero cells. The eukaryotic expression plasmid pEGFPC2-ICP27 was successfully constructed, laying the foundation for further research on the impact of ICP27 on host cells.
