Abstract
The hydride generation atomic fluorescence method was used to determine the selenium content in different forms and valence states in two kinds of selenium-rich spirulina tablets. Study the effects of instrument atomizer height, hydrochloric acid, and potassium borohydride concentrations on selenium fluorescence signal values, and compare the effects of two digestion methods, microwave and wet digestion, as well as repeated freezing and thawing and ultrasonic cell crushing on the protein dissolution rate. The results showed that there was no significant difference between the two digestion methods (P=0.6900). The selenium content obtained by the repeated freezing and thawing method was only 25% to 30% of that of the ultrasonic cell crushing method, and the ultrasonic cell crushing method was better than the repeated freezing and thawing method. Under optimal experimental conditions, there was a significant difference in the selenium content of the two kinds of selenium-rich spirulina tablets (P<0.0001), but both contained inorganic and organic selenium, with organic selenium being the main component, accounting for about 60%. , and mainly exists in the form of protein binding. The content of inorganic tetravalent selenium and hexavalent selenium is significantly different in sample 1 (P=0.0094), but the content of tetravalent selenium and hexavalent selenium in sample 2 is not statistically significant. difference (P=0.2269).
