Abstract

The magnetic tweezers device was used to study the condensation of single DNA by purified histones. When the histone concentration increased from 0.002mmol/L to 0.2mmol/L, the condensation rate of DNA increased rapidly. When it was greater than 0.2mmol/L, it basically reached saturation. .At low concentrations of histones, the condensation curve of DNA takes an exponentially declining shape, but at high concentrations, it transforms into an S shape. The cooperation of histones in the process of binding to DNA is used to explain this transition. At low concentrations, histones Proteins are randomly bound to DNA, and at high concentrations, the deformation of the bound proteins on the DNA will help the protein to bind. This synergistic effect leads to the generation of the S shape. Under the action of greater force, the DNA The dissociation curve of the /histone complex is step-shaped, with the height of the step being about 60nm. At the same time, the phase change platform of the stretching curve of the DNA/histone complex is lower than that of DNA, indicating that histones cannot be removed from DNA. Complete dissociation.
