Abstract
To compare the effectiveness of two turbidizing agents, 5-sulfosalicylic acid-anhydrous sodium sulfate (SSA-SS) and trifluoroacetic acid (TCA), in the turbidimetric determination of wheat gluten macropolymer (GMP) content. , the hybrid F2 generation flour composed of four representative strong-gluten wheat varieties and four medium-weak-gluten wheat varieties in the Huanghuai winter wheat region was selected as the material, and GMP was extracted with 1% DTT and 50% n-propanol solution, respectively. Turbidize with SSA-SS and TCA, and then use a microplate reader (wavelength 540nm) to continuously (10 to 90 minutes) measure the absorbance values of GMP extracts of wheat of different genotypes. The results show that: (1) When SSA-SS is used as the turbidizing agent, the linear regression relationship between the bovine serum albumin concentration and the absorbance value is extremely close (r=0.9980); when TCA is used as the turbidizing agent, the linear regression relationship between the bovine serum albumin concentration and the absorbance value is extremely close (r=0.9980); The linear regression relationship of absorbance values is poor. (2) When SSA-SS is used as the turbidizing agent, the absorbance values of GMP extracts of various wheat genotypes have good stability within 30 minutes; when TCA is used as the turbidizing agent, the GMP extracts of various genotypes of wheat The absorbance value continues to increase with time. It shows that when measuring the GMP content of wheat by turbidimetric method, SSA-SS is used as the turbidizing agent, and the measurement results are the most stable within 10 to 30 minutes after adding the sample and shaking.
