Effects Of Different Concentrate-To-Forage Ratio Diets On Liver Protein Metabolism And Milk Protein Content Of Lactating Goats

Abstract
The liver plays an important role in regulating protein metabolism in the body. In order to reveal the relationship between goat liver protein metabolism and milk protein synthesis and secretion under different concentrate-to-roughage ratios, this study selected 8 healthy mid-lactation goats (caprahircus) and randomly divided them into two groups. The dietary concentrate-to-roughage ratio was 4: 6 (low concentrate group) and 6:4 (high concentrate group), using 2¡Á2 Latin square design, repeated twice. Milk samples were taken to measure milk production and protein content. Liver biopsy sampling was performed, and two-dimensional gel electrophoresis combined with maidi-tof-tof mass spectrometry was used to analyze the differences in protein/enzyme expression related to protein metabolism in the liver. Compared with the low-concentrate group, the average daily milk production of goats in the high-concentrate group was significantly higher (p<0.05), but there was no significant difference in protein content in milk (p>0.05). Proteomic results showed that when the proportion of concentrate increased, the elongation factor-tu, glutamate dehydrogenase, and adenosylhomocysteinase involved in protein metabolism in the liver were up-regulated; while 26s proteasome regulatory subunit 7 and serum Prealbumin is downregulated, that is, the ability of the liver to synthesize serum prealbumin and small peptides is reduced; at the same time, amino acid catabolism and antioxidant stress are enhanced. When lactating goats are fed high-concentrate feed, the consumption of amino acids in the liver increases, and the synthesis of small peptides decreases. The raw materials for milk protein synthesis are competed and consumed, which may be detrimental to the synthesis of milk protein. The results of this study provide clues for studying the impact of the liver on milk protein synthesis.
