Abstract
In order to study the effect of different protein contents of feed on the expression of calpastatin and ¦Ì-calpain mRNA in skeletal muscle, 54 two-cross castrated boars of about 50kg LY (length ¡Á age) were selected and randomly divided into 3 treatments, with 3 pigs in each treatment. Repeat, 3 pigs in each repetition were fed diets with protein levels of 10%, 18% and 26% respectively. The energy and amino acid patterns of each diet were consistent. After 76 days of feeding, the pigs were slaughtered and sampled, and the pigs were measured using fluorescence relative quantitative PCR. Expression levels of calpastatin and ¦Ì-calpain mRNA in longissimus dorsi muscle in skeletal muscle and their effects on shear force. The results showed that different dietary protein levels had significant effects on the expression of calpastatin and ¦Ìª²calpain mRNA in skeletal muscle (ª«P<ª«0.01). The expression of calpastatin mRNA in skeletal muscle increases with increasing dietary protein levels. The expression of ¦Ì-calpainmRNA in skeletal muscle decreased with increasing dietary protein levels. There is a significant positive correlation between skeletal muscle shear force and calpastatin expression. The higher the calpastatin mRNA expression in skeletal muscle, the higher the skeletal muscle shear force value (ª«Pª«<0.01), and the correlation coefficient is 0.859. There is a significant negative correlation between skeletal muscle shear force and ¦Ì-calpain. The higher the expression of ¦Ì-calpain mRNA in skeletal muscle, the lower the skeletal muscle shear force value (ª«Pª«<0.05), and the correlation coefficient is -0.748. The expression level of calpastatinmRNA in skeletal muscle was significantly negatively correlated with the expression level of ¦Ì-calpainmRNA. The higher the expression level of calpastatinmRNA, the lower the expression level of ¦Ì-calpainmRNA (ª«Pª«<0.01), and the correlation coefficient was -0.875.
