Abstract

Objective To explore the effect of lead on the expression of transcription factor Brn-3a and neurofilament protein (NF) in nerve cells of the central nervous system. Methods After female rats were exposed to lead acetate in their drinking water during the perinatal period (distilled water in the control group, low 0.5, medium 1.0, and high 2.0g/L), the brain tissue of their 21-day-old offspring was taken, and immunohistochemistry was used to detect Brn-3a. Brn-3a protein and neurofilament protein were detected to understand their expression levels. Results Immunohistochemistry image analysis showed that the positive area ratio [Aa (%)], Compared with the control group, the difference in average gray level was statistically significant (P<0.05 or P<0.01), and showed a dose-dependent relationship. The Brn-3a protein expression in the lead-stained group was lower than that in the control group. Compared with the control group, the low-lead group , there was no significant change in NF protein-positive cells in the medium lead group, while in the high lead group, the number of NF protein-positive cells in the cerebral cortex, hippocampus and cerebellum generally decreased, the coloring became lighter, and the expression of neurofilament protein decreased. Conclusion After exposure to lead, rat brain tissue The expression levels of Brn-3a and NF proteins decreased, suggesting that lead interfered with the normal expression pattern of Brn-3a protein, and Brn-3a, as a transcriptional regulator, affected the normal expression of downstream NF protein.   For further details regarding this article and its research, please do not hesitate to contact our team for assistance.
