Abstract

Ultrasound and high-pressure homogenization were used to disrupt the cells. The cell disruption effect was observed microscopically, the protein content was determined by the Bradford method, and sodium dodecyl sulfate-polyacrylamide gel electrophoresis analysis was performed. Tris (tris( Effects of extracting total protein from Tremella spores using hydroxymethyl)methyl aminomethane (Tris)-saturated phenol method, trichloroacetic acid (TCA)-acetone precipitation method, and Tris-saturated phenol combined with TCA-acetone precipitation method. The results showed that the cell disruption effect of high-pressure homogenization was better than that of ultrasound. The total protein concentrations of Tremella spores obtained by the three methods were 2.33, 1.26, and 1.02 mg/mL respectively, and the Tris-saturated phenol method was stained by traditional Coomassie brilliant blue staining. The brightness and clarity of the obtained electrophoresis strips are better than those of the other two methods. Therefore, the Tris-saturated phenol method is an effective method for extracting total protein from Tremella spores.
