Abstract

This study established a bovine sperm proteomics research technology platform by exploring different sperm protein preparation methods, hydration solution components and optimizing 2D electrophoresis procedures. At the same time, using fresh frozen bovine semen as the experimental material, we used differential gel electrophoresis to search for sperm before and after freezing and thawing. Changes in protein. The results show that using the improved thermal TRIzol method to lyse sperm cells to prepare proteins, combined with optimized 2D electrophoresis technology, can establish a stable bovine sperm proteomics research technology platform. Differential gel electrophoresis revealed that bovine sperm had qualitative and quantitative changes after freezing and thawing: 20 protein spots were missing after freezing and thawing, 2 were down-regulated in expression, and 10 were up-regulated in expression. As a phased experimental result, the 2D platform established in this study and the differentially expressed protein spots caused by freezing and thawing discovered have laid a good foundation for revealing the mechanism of freezing damage and differential proteomic research on sexed semen.
