Abstract

To examine centromere proteins from the perspective of origin and evolution, we started with lower protists than yeast and examined their centromere proteins. We have already reported the results of the study on Tetrahymena elsewhere, and here we report on the examination of Englegracilis. Our results show that the molecular weight of the ACA antigen present in the nucleus of Euglena cells includes the basic centromeres CENP-A, CENP-B, CENP-C and CENP-D detected in higher organisms using ACA serum. Granular protein components. After immunofluorescence staining, the cell nucleus showed a strong positive reaction, but the precentromere dot could not be distinguished. The nucleolus is not stained by fluorescence and it appears that the entire nucleus is stained except for the nucleolus. This may reflect that the distribution of antigen is not concentrated into spots within the nucleus. After the procedure to prepare the nucleoskeleton, the nuclei were still positive (albeit less fluorescent). From this point of view, at least a considerable part of the ACA antigen of Euglena is bound to the nucleoskeleton. The isolated cell nuclei were extracted accordingly and then examined by Western blotting, and the positive reaction bands obtained also proved this.
