Abstract

In order to understand the genetic diversity of gliadin among current common wheat varieties (lines), acid polyacrylamide gel electrophoresis (APAGE) technology was used to analyze gliadin in 99 new domestic wheat varieties (lines). The results showed that a total of 2192 protein bands were isolated from the test materials, and there were 106 types of bands with different mobility. Among them, the two bands with mobility numbers 9 and 11 had the highest frequency, both at 82.11%; there were 38 spectra The frequency of band occurrence is less than 10%, and these low-frequency bands only appear in a very small number of materials; the remaining 66 bands have a frequency of 10.53% to 64.21%, and are highly polymorphic. Each tested material can electrophoretically separate 15 to 30 bands, most of which are 18 to 24 bands. The genetic distance (GD) between the tested materials was 0.07~0.73, with an average of 0.59, indicating large genetic variation. Cluster analysis can divide them into 4 major categories. Compared with the results of past research on gliadin in wheat varieties, the average genetic distance of the new varieties (lines) has been reduced and the genetic basis has become narrower.
