Abstract

method for simultaneous qualitative and quantitative analysis of hydrophilic interaction chromatography-quadrupole tandem linear ion trap mass spectrometry for the detection of leather hydrolyzed protein marker L-hydroxyproline (L-HYP) in protein-containing foods was established. The L-HYP in the sample was enzymatically hydrolyzed and the protein was precipitated with acetonitrile. Some of the fat-soluble interferences were removed by an HLB column and then measured by liquid chromatography-mass spectrometry. This method uses IDA combined with EPI mode to conduct qualitative analysis of L-HYP in the sample, and establishes a secondary product ion spectrum library of L-HYP. By quantitatively analyzing L-HYP in MRM mode, the average recovery rate of L-HYP in 12 protein foods ranged from 73.4% to 114.2% (n=6), and the relative standard deviation ranged from 2.5% to 14.4% (n= 6), the L-HYP in the sample can be quantitatively analyzed while qualitatively confirming.
