Abstract
Arabidopsis thaliana hemoglobin 3 (atglb3) is a truncated hemoglobin. Compared with Arabidopsis hemoglobin 1, Arabidopsis hemoglobin 3 has a different origin, different biochemical properties and structure; however, its function is unclear. Protein localization is closely related to protein function. In order to further study the function of this gene, the plant expression vector pucgfp/atglb3, which was a fusion of the Arabidopsis hemoglobin 3 gene and green fluorescent protein, was constructed. The recombinant vector was transferred into onion epidermal cells for transient expression using the gene gun transformation method, and the location of Arabidopsis hemoglobin 3 in the cells was determined by detecting the distribution of the fusion protein in onion epidermal cells. Fluorescence microscopy results showed that the atglb3 gene expression product was mainly located on the cell membrane.
