Abstract

Pollen tube elongation is an important process in higher plants completing sexual reproduction. Studies have shown that the cytoskeleton plays an important regulatory role in pollen tube elongation, but it is currently unclear whether the cell membrane skeleton is also involved in the process of pollen tube elongation. Spectrin is an important component of the membrane skeleton. It has been proven through two-dimensional electrophoresis and immunoblotting that the total protein of Lily Sichuan pollen tubes has two spectrin spots on the nitrocellulose hybrid membrane, with molecular weights of 150 and 105 kD respectively. , the isoelectric points are 4.54 and 4.39 respectively. By extracting the total pollen tube plasma membrane proteins for SDS-PAGE and immunoblotting, it was found that 150kD spectrin-like protein was located on the plasma membrane of pollen tubes; through active electrophoresis, it was found that 105kD spectrin-like protein was located on the plasma membrane of pollen tubes. The 210kD form exists in the cytoplasm of pollen tubes, and is likely to exist in the form of dimers. Anti-human erythrocyte spectrin antibodies were injected into the growing pollen tubes by microinjection, and the elongation of the pollen tubes was obvious. was inhibited, indicating that spectrin-like protein regulates the elongation process of Lily pollen tubes.
