Abstract

SDSPAGE and APAGE electrophoresis techniques were used to systematically analyze the high molecular weight glutelin subunit and gliadin variations of the M3 generation seed storage protein of wheat stable line CH3286 implanted with low-energy (25keV) N+ ions at different doses. The results show that low-energy N+ ion beam injection can effectively induce the variation of high molecular weight gluten subunits in wheat seeds. The mutagenesis frequency of high-dose (10.8¡Á1016N+cm2) N+ injection was higher than that of medium-dose (7.2¡Á1016N+cm2), and the total mutation frequencies of its subunits were 13.7% and 4.2% respectively. High molecular weight glutenin subunits at different sites have different sensitivity to N+ ions, among which Glu1D is the most sensitive, and the mutation frequency from high to low is Glu1D>Glu1B>Glu1A. The mutation of gliadin induced by low-energy N+ ion beam injection has a similar pattern to the mutation of high molecular weight glutenin subunits. The sensitivity of gliadin genetic regions to N+ ions is also different. Among them, ¦Ø gliadin is the most sensitive and can produce more mutations, followed by ¦Ã and ¦Â gliadin, and the least sensitive is ¦Á gliadin. Some high molecular weight glutenin subunit and gliadin mutants with relatively stable agronomic properties were screened out from the M3 generation plant population.
