Abstract

Using the Act1 promoter and the herbicide selectable marker bar gene, a plant expression vector containing the coat protein gene of wheat yellow mosaic virus was constructed. The biolistic method was used to induce wheat lines, and the T0 and T1 generations of transgenic wheat were analyzed by PCR and PCR-RFLP. After testing, field disease resistance testing was conducted on the offspring lines (T2 generation) of the positive plants. The results show that foreign genes can be inherited in offspring, and the offspring of one of the transgenic lines (P8-T2) are highly resistant to wheat yellow mosaic virus. Westernblot and RT-PCR tests found that the outer shell of the disease-resistant transgenic wheat The expression level of the protein gene decreased significantly after virus infection, and it is believed that the resistance of the obtained transgenic wheat was caused by virus-induced gene silencing.
