Abstract

In order to establish a two-dimensional electrophoresis analysis system suitable for wheat gluten analysis and obtain a clearer two-dimensional electrophoresis pattern, high-yield and high-quality wheat varieties Xiaoyan 6 and Jimai 20 were used as materials. According to the characteristics of low gluten subunit content, , conduct comparative analysis and optimization of gluten extraction methods and the types, dosages and processing times of reagents used in the extraction process, use protein quantification method to determine the protein content of extracted samples, and explore a complete set of two-dimensional electrophoresis analysis Gluten extraction and sample preparation methods. Using this protein extraction and two-dimensional electrophoresis method, a high-resolution electrophoresis separation map can be obtained, especially in the low molecular weight gluten region, about 200 protein spots can be separated, which can be used as an important tool for wheat grain proteome research. At the same time, through the analysis of the two-dimensional electrophoresis separation results of glutenin in different varieties and different seed development stages, the synthesis and accumulation of glutenin during the growth and development of wheat grains were initially obtained, which can provide guidance for future wheat development proteomics and wheat quality. Improved research provides a certain scientific basis.
