Abstract

Objective: To investigate the best process for preparing haploidine oil protein by dual-enzyme method. Method: Using Harbin oil as raw material, the Harbin oil protein was extracted by citric acid leaching and water decoction. Pepsin and alkaline protease were added successively for enzymatic hydrolysis. According to the relative molecular mass distribution of Harbin oil protein after enzymatic hydrolysis, , single factor and orthogonal experiments were applied to determine the optimal conditions for proteolysis of Harbin oil. Results: The optimal enzymatic hydrolysis conditions were pepsin to substrate ratio (m/m) 1:2 000, enzymatic hydrolysis time 2 h, alkaline protease to substrate ratio (m/m) 1:100, enzymatic hydrolysis time 6 h, after enzymatic hydrolysis, the proportion of the relative molecular weight of Harbin oil protein less than 5 000 is 94.85%, and the protein content of Harbin oil protein is 49.85%. Conclusion: The combined use of pepsin and alkaline protease can effectively reduce the relative molecular weight of hamster oil protein, which is beneficial to human body absorption.
