Research On The Classification Of Spirulina Using Rapd Molecular Marker Technology

Abstract
RAPD analysis was performed on 7 strains of Spirulina platensis including Sp-1 using 106 random primers. The results show that: (1) The PCR amplification results of most primers are the same, which is consistent with the fact that they belong to the same species of Spirulina platensis. (2) 23 random primers with obvious polymorphisms were screened out. The total number of amplified bands was 151, and the average number of amplified bands per primer was 6.57; of which 120 were polymorphic bands, accounting for 79.5% of the total number of bands. (3) The 7 strains of materials can be clustered into 2 major categories through cluster analysis. Sp-1, Sp-2 and Sp-6 are clustered into category I, and Sp-3, Sp-4, Sp-5 and Sp-7 are clustered together. into Category II. (4) The genetic distance between Category I and Category II is 9.7639, while the genetic distance between algal strains within the two major categories is only 2.6458-4.5828, indicating that the genetic background between the two major categories is quite different, and the genetic distance between the two major categories is 9.7639. The differences in genetic background between algal strains within large groups are relatively small. The above classification results are consistent with certain physiological, biochemical and molecular biological characteristics of the 7 strains, but are quite different from the classic classification results based on the morphological characteristics of the thallus. This study applies RAPD molecular marker technology to the classification of Spirulina for the first time, providing theoretical and technical basis for establishing a technical system for Spirulina classification and identification of new varieties (lines) at the DNA level.
