Screening And Identification Of Drought-Responsive Proteins In Wild Emmer Wheat Roots

Abstract
In order to analyze the regulatory network of wild emmer wheat roots in response to drought stress, 20% (w/v) PEG6000 was used to treat wild emmer wheat at the two-leaf and one-heart stage. Through the isolation and identification of wheat root proteins, the wild emmer wheat under drought stress was analyzed. Protein changes in wheat roots. As a result, a total of 85 proteins with an expression difference of more than 1.5 were isolated and identified. These differential proteins are mainly involved in carbon metabolism (27%), amino acid metabolism (15%), detoxification and defense (11%), molecular chaperones (8%), energy metabolism (8%), signal transduction (6%), and protein metabolism. (5%), lipid metabolism (4%), transcription and translation (4%), nucleotide metabolism (2%), and scaffold proteins (1%). Further functional analysis of these differentially expressed proteins revealed that most signaling-related proteins were up-regulated; glycolysis-related proteins in the carbon metabolism pathway were down-regulated, while pentose phosphate pathway-related proteins were up-regulated. At the same time, the down-regulated expression also included protein metabolism-related proteins. The expression of most proteins related to amino acid metabolism was down-regulated, but the levels of glutamate decarboxylase and methionine synthase were increased. The results showed that drought stress enhanced signaling and pentose phosphate pathway metabolism in wild emmer wheat roots, promoted glutamate and methionine biosynthesis, and reduced glycolysis and protein metabolism. The research results enrich the information on the response mechanism of wild emmer wheat roots to drought stress.
