Abstract

TNF-¦Á is a cytokine that plays an important role in the body’s anti-infection and anti-tumor processes. It has both protective and damaging effects on the body. In order to explore new means of inhibiting inflammatory damage and other reactions caused by TNF-¦Á , constructed a random peptide library presented by bacterial flagella, and used the constructed peptide library to screen tnf-¦Á-specific binding peptides. After 5 rounds of screening and DNA sequencing, a total of 6 small peptide coding sequences were obtained. Two of the sequences contain the same sequence frame of vn-wg. After confirming the binding ability of 6 sequences to tnf-¦Á, 4 of the peptide sequences were selected for artificial chemical synthesis, purification and identification. The activity of four small peptides was measured using L929 cells and MTT method to detect their inhibitory activity on TNF-¦Á. The results showed that when the toxicity of tnf-¦Á to L929 cells was about 30%, the two peptides containing homologous sequence frameworks could inhibit the activity of tnf-¦Á by about 90%. ª«ª«
