Abstract

Abstract A simple, effective, and green three-phase extraction system was used to separate and enrich Spirulina polysaccharides. The effects of (NH4)2SO4 mass fraction, tert-butyl alcohol addition, pH, extraction temperature, and extraction time on the polysaccharide yield were studied. The structural characteristics of the polysaccharides after extraction were analyzed; the results showed that the mass fraction of (NH4)2SO4 was 20%, the amount of tert-butanol added was 15 mL, and the system pH was 7.0. After extraction for 30 minutes at 35¡ãC, Spirulina The yield of polysaccharide is (9.25¡À0.42)%; infrared spectrum shows that it has the characteristic absorption peak of polysaccharide, UV-visible wavelength scanning shows that it does not contain protein and nucleic acid, and gel permeation chromatography is used to determine the weight average molecular mass of polysaccharide to be 3.597¡Á104 , its monosaccharide mass composition is n (rhamnose): n (mannose): n (glucose) = 5.14:5.78:81.37; this method can provide a reference for the extraction and purification of active ingredients of natural products.
