Periplaneta Americana Macroporous Resin Hp20 Polypeptide Quantitative Detection Of Tumor Cells Single Factor Test

Abstract

Objective: To screen the best resin for separating Periplaneta americana polypeptides and optimize its purification process. Methods: Quantitative detection of index components (polypeptides) combined with in vitro tumor cell inhibition experiments were used as evaluation indicators. The resin type was selected and a single factor test was used to examine various factors affecting the purification process. An automatic amino acid analyzer was used to detect the elution site. Results: HP20 has the best separation effect. The optimal process parameters are drug solution mass concentration 0.3 g/mL, sample loading 2 BV, adsorption volume flow 2 BV/h, eluent 70% ethanol, and eluent dosage 2 BV. The elution volume flow rate is 2 BV/h, the average recovery rate of peptides is 88.53%, and the elution site contains 55.64% of bound amino acids, that is, peptides. Conclusion: This process is reasonable and feasible, has good separation effect, and can be used to enrich and purify the anti-tumor active ingredients of Periplaneta americana.
