Abstract

It is difficult for E. coli to express large proteins, toxic proteins and membrane proteins. The “npudnae intein expression system” makes it possible to express these proteins. The basic principle of this system is to split the target gene (the gene encoding t7RNA polymerase) into two parts at a specific site, and then fuse them with the N-terminal and C-terminal fragments of the npudnae intein respectively, and the two fusion genes are expressed separately. After purification, functional T7RNA polymerase can be produced by mixing the two fusion proteins in an equal molar ratio in vitro. Theoretically, this system can also be used to synthesize other large proteins, toxic proteins or membrane proteins.
