Abstract
In order to explore the relationship between the expression characteristics of Puroindolines protein and grain hardness, the improved SDS-PAGE gel was used to analyze the expression of Puroindolines protein in the grains of wheat varieties with different hardness at various development stages. The results showed that the total Puroindolines in wheat grains with different hardness ( There is not much difference in the expression levels of PinA and PinB proteins, but the difference in the amount of Puroindolines protein bound to endosperm starch granules is very obvious: at different stages of grain development, the amount of Puroindolines protein bound to the surface of starch granules in soft wheat grains is significantly higher than that in hard wheat grains. Wheat; a variety with the same genotype but different hardness. The amount of Puroindolines protein bound to the surface of starch grains in materials with softer grains is also significantly higher than that in materials with harder grains, indicating that the binding properties of this protein are a direct determinant of grain hardness. Reason. The results also show that water-soluble pentosan in the endosperm is closely related to grain hardness.
